摘  要:目的:探讨人胎儿毛乳头细胞与生物可降解材料聚羟基丁酯的细胞相容性,为进一步以聚羟基丁酯为基质用人胎儿毛乳头细胞诱导表皮干细胞分化为毛囊,从而构建具有毛囊结构的组织工程皮肤提供实验依据和理论基础。方法:实验于2004-03/07在第四军医大学附属西京医院烧伤外科实验室完成。标本为5~7个月胎龄终止妊娠人流胎儿(家属知情同意)。将二步法培养的人胎儿毛乳头细胞接种于聚羟基丁酯浸泡液及膜片上,四甲基偶氮唑盐法测定细胞黏附率和细胞增殖指数,并在体外培养1,4d后扫描电镜观察细胞的生长状况。结果:①人胎儿毛乳头细胞在聚羟基丁酯浸泡培养基中生长良好。②24h,48h,72h四甲基偶氮唑盐法结果与对照组无显著差异,4h后人胎儿毛乳头细胞在聚羟基丁酯支架上黏附率为79.06%。③电镜显示人胎儿毛乳头细胞生长良好,形态正常,黏附牢固。结论:人胎儿毛乳头细胞与聚羟基丁酯相容性,黏附性良好,可以用于进一步构建具有毛囊结构的组织工程皮肤。AIM: To study the cyto-compatibility between human fetal hair papilla cells and polyhydroxybutyrate (PHB), in order to provide experimental evidence and theoretical foundation for fabricating tissue engineering skin with hair follicle differentiated by epidermal stem cells induced by human fetal hair papilla cells using PHB as base. METHODS: The experiment was conducted at the laboratory of Department of Bums, Xijing Hospital, Fourth Military Medical University of Chinese PLA between March and July 2004. Fetuses of artificial abortion that was termination of pregnancy, aged 5-7 months, were selected with the agreement of the family member. Human fetal hair papilla ceils cultured with double footwork were inoculated at soak fluid and chaff of PHB. Adherence rate of cells and proliferation index were detected with MTY assay, and growing condition of cells was observed with scanning electron microscope (SEM) after cultured for 1 and 4 days in vitro. RESULTS: ① Human fetal hair papilla cells grew well in PHB medium. ②There was insitoificant difference between the result of 24 hours, 48 hours and 72 hours MTT assay and that in the control group. The adherence rate of human fetal hair papilla cells in PHB carriage was 79.06%. ③ Electron microscope showed that human fetal hair papilla cells was growth well, eumorphism firm adherence. CONCLUSION: The human fetal hair papilla cells have good compatibility with PHB, having good adherence, and can be used to build further tissue engineering skin with hair follicle structure.
