摘  要:目的探讨在建立抗鼠疫F1抗原的单克隆抗体杂交瘤株的过程中抗原免疫的剂量及免疫方法.方法采用F1抗原常规免疫、改良的F1抗原脾内注射免疫以及EV菌免疫3种方法免疫6~8周龄,体重为10~20g,健康的雌性BALB/c小鼠,并用间接ELISA法检测尾血中的抗体. 结果在常规免疫中,200μg 组和100μg组免疫效果优于50μg组,而200μg组和100μg组免疫效果基本一致;改良的F1抗原脾内注射免疫,血中抗体效价较高;EV菌免疫的效果不理想. 结论用F1抗原常规免疫小鼠,免疫剂量用100μg/只较为合适;若要缩短免疫时间,采用改良的脾内注射免疫法是理想的选择.Objective To investigate the immne doses and methods of F1 antigen during the establishment of hybridoma cell strains secreting McAbs against F1 antigen of Y. Pestis. Methods Healthy female BALB/c mice of six to eight weeks old and 10 -20 gram weight, were immunized with three methods, F1 antigen conventional immunization, injecting immunization via spleen with F1 antigen and immunization of EV bacterium via abdominal cavity. Blood collected from the mice was tested for anti -F1 antibody titres by indirect enzyme - linked immunosorbent assay (ELISA). Results Among three immune methods, as for conventional immunization, the immune effect of 200μg group and 100μg group was better than that of 50μg group, while the immune effect of 200μg group and lO01xg group was almost the same; the titres of injecting immunization via spleen with F1 antigen was higher than that of another two methods ; the immune effect of EV bacterium was not good. Conclusions The immune dose of 100μg for mouse is appropriate with FI antigen conventional immunization ; injecting immunization via spleen would be a good choice if researcher want to shorten the immune time.
