摘  要:目的:探索一种简便易行的伊氏锥虫(Trypanosomaevansi)低温冷冻保种新方法。方法:感染伊氏锥虫小鼠的肝和脾组织分别冻存于-20℃和-80℃低温冰箱7、10、60d,观察复苏后伊氏锥虫的形态、活力及对小鼠的感染力。结果:伊氏锥虫在-20℃和-80℃温度下冻存7、10、60d后,光镜下形态与新鲜组织内虫体一致。随着冻存时间延长虫体活力稍有下降。肝、脾组织-20℃或-80℃冻存60d后虫体活力分别为74.47%,74.43%和74.62%,74.96%,差异无统计学意义(P>0.05)。经-20℃和-80℃冷藏保存7、10d后接种,小鼠虫血症出现时间均为5d,保存60d者为7d。两种温度,不同时间保存虫体复苏后对小鼠感染成功率和致死率均为100%。结论:不加保护剂直接冷冻受染小鼠肝或脾组织保存伊氏锥虫,不失为一种经济、便捷的保种方法。Objective:To explore a convenient freezing method to preserve Trypanosoma evansi. Methods: The liver and spleen fractions of mice infected by trypanosoma evansi were stored at -20℃ and -80℃ in ordinary refrigerator or deep freezer for 7,10 and 60 days,respectively. The resuscitative trypanosoma evansi was investigated in morphology and infectivity. Result.Compared with parasites from fresh tissues there were no significant changes in morphology. The viability of organisms kept decreasing slowly when the freezing time went longer. The viabilities of organisms in the liver and spleen stored at -20 ℃and - 80℃ for 60 days were 74.47%,74.43% and 74.62%, 74.96%, respectively, showing no significant difference among these groups ( P 〉0.05). All the latent periods of parasitemiae in mice infected with parasites stored for 7 or 10 days were 5 day, and those of 60 day storage at both -20℃ and -80℃ were 7 day. All the positive infecting rates and death rates of mice infected by freezing kept trypanosoma evansi were 100%. Conclusion:It is an economical and convenient way to store trypanosoma evansi by directly freezing the infected mouse liver or spleen without any protective reagents.
