摘  要:目的:测定银黄颗粒剂中黄芩苷在大鼠血浆中的浓度及其药代动力学,为临床合理用药提供科学依据.方法:采用高效液相色谱法,0DS C18柱(4.6 mm×250 mm,5μm),流动相:甲醇-磷酸二氢钠缓冲液(pH:2.7,54:46),检测波长:276nm,流速:1.0ml·min-1,柱温:40℃.结果:黄芩苷在0.05~10μg·ml-1范围呈良好的线性关系,回归方程为:Y=0.6575X-0.1479,r2=0.9996,当信噪比(S/N)≥3时,血浆中黄芩苷最低检测浓度为8 ng·ml-1.黄芩苷在大鼠体内呈二室模型分布,其主要药动学参数为Vd:2.65 L,K12:5.36 h-1,K21:0.71 h-1,Ke:4.25h-1,t1/2:0.17 h,AUC:2.01μg·ml-1·h.结论:本方法简单、灵敏、准确,可用于黄芩苷血药浓度分析及其药代动力学研究.Objective: To examine the concentration of baicalin of Yinhuang granules in big rat internal body and its pharmacokinetics, supplying scientific accords for using drugs reasonably in clinic. Method: To adopt the HPLC, ODS,s Column (4.6mm×250mm, 5μm), mobile phase: methanol-natrium biphosphoricum balanced solution (pH : 2.7,54 : 46 ) , detection wavelength : 276nm, flow rate:1.0ml. min^-1, temperature: 40℃. Result: baicalin showed a good linearity in 0.05-10μg. ml^-1, regression equation:Y=0.6575X-0.1479,r^2=0.9996, when S/N≥3, the lowest detection concentration of baicalin was 8 ng·ml^-1 in blood plasma, baicalin took on the two-compartment model in big rat internal body. Its important pharmacokinetics parameter: Vd :2.65 L, K12:5.36 h^-1 ,K21:0.71 h^-,Ke:4.25h^-1 ,t1/2:0.17h, AUC:2.01μg. ml^-1. h. Conclusion: The method is simple, accurate, sensitiveness, can be used to analyze baicalin concentration in blood of and pharmacokinetics.
