摘　　要：在pH 3.80～4.20的Clark-Lubs缓冲介质中,蛋白质与Cu(Ⅱ)-ARS络合物作用,其吸收光谱发生大幅度变化,生成的深红色三元复合物,λmax为538nm,比试剂本身红移118 nm,比络合物λmax红移30 nm.研究了有生物大分子参与的三元复合物反应体系的光谱性质及其影响因素,解释了初步机理,在确定的最佳实验条件下,建立了以Cu(Ⅱ)-ARS络合物为光谱探针光度法测定人血清蛋白质的新方法.蛋白质在0～300mg/L的范围内符合比尔定律,表观摩尔吸光系数ε538=2.37×105L·mol-1·cm-1(人血清白蛋白).用于实际人血清样品的测定基本无干扰,灵敏度比茜素红S探针法高23.7倍.Copper（Ⅱ） -Alizarin red S complex couht combine with protein to form a deep red ternary complex in Clark - lubs at pH 3.8 - 4.2. The complex has a maximum absorption at 538 nm. The spectral characteristic of the reactive system of ternary complex and the influencing factors were studied. A new method for the determination of protein in HAS by using Copper（Ⅱ） - Alizarin red S complex as a spectral probe was established under optimum conditions. Beer＇s law is obeyed in the concentration range of 0-300 mg/L and the apparent molar absorptivity is 2.37×10^5 L·mol^-1·cm^-1. The method has been applied for the determination of total amounts of proteins in the human serum samples with satisfactory results.
