摘 要:目的:建立测定云南常用药材雪上一枝蒿(Aconitum brachypodum Diels.)中雪上一枝蒿甲素的反相高效液相色谱法,考察3种不同来源的雪上一枝蒿中雪上一枝蒿甲素的含量。方法:采用 Extend-C_(18)色谱柱(250 mm×4.6 mm,5 μm);磷酸盐缓冲溶液(pH 7.3)-甲醇(28:72)为流动相;检测波长为210nm。结果:雪上一枝蒿中雪上一枝蒿甲素成分与其他成分能得到很好的分离,不同产地雪上一枝蒿药材中雪上一枝蒿甲素含量差异较大,以云南东川产的雪上一枝蒿甲素含量最高。雪上一枝蒿甲素平均回收率为98.4%,RSD 为1.7%(n=5)。结论:该方法便捷、灵敏、准确,重复性好,可以控制该药材的质量。Objective:To establish an RP - HPLC method for determination of bullatine A in Aconitum brachypodum Diels. which commonly used in traditional medicine of Yunnan. To check the content of bullatine A in Aconitum brachypodum Diels. from different origins. Method:Using an Extend - C 18 column (250 mm 4. 6 mm,5 μm). A mobile phase consisted of phosphate buffer( pH 7.3 ) - methanol(28: 72)was used. The flow rate was 1.0 mL · min^-1, and the UV detector wavelength was 210 nm. Results:Bullatine A was well separated from other comnounds of Aconitum brachypodum Diels. The contents of bullatine A in Aconitum brachypodum Diels. was more different in differenting areas. The contents of bullatine A in Aconitum brachypodum Diels from Dongchuan,Yunnan was higher than that of from others areas. The average recovery of bullatine A was 98.4% and RSD was 1.7% (n =5). Conclusion:The method is accurate and reliable,suitable for bullatine A in Aconitum brachypodum Diels.
