A PRELIMINARY STUDY ON THE MATERIAL RESOURCE OF DNA IN THE DNA FINGERPRINTING ANALYSIS OF GIANT PANDAS

摘  要:以圈养大熊猫4个家系,13只个体的31份粪便、尿液以及甲醛固定的睾丸组织等作材料,辅以血液、被毛进行DNA指纹分析。结果表明,同一个体的血液、被毛、粪便、尿液,以及甲醛固定的睾丸组织材料的DNA指纹图谱完全一致.使用两种寡核苷酸探针重复验证圈养大熊猫5胎6仔的父子关系,效果稳定。DNA were isolated from fomalin-fixed testes, excrements, urine, blood and hairs of 13 giant pandas in 4 families in Chensdu Zoo and Chengdu Research Base of Giant Panda Breeding.Using the method of DNA fingerprinting we made an analysis of the DNA fingerprints to the 4 families, and obtained the following results:1. The DNA fingerprints of blood, hairs, urine, formalin-fixed testes and excre ments of the same giant panda are identical. It indicates that all the materials above are the resource of DNA in the research works of DNA fingerprinting analysis of giant panda.2. Both of the oligonucleotide probes of JH 12. 6 provided by Jinhe Applied Biotechnology Institute and LZF-1 made by Molecular Biology Institute of Sichuan Normal University can form the hybrid molecules with the simple repetitive DNA of genome of giant pandas, and all of the hybrid bands of any filial generation farrowed down by monogamy and polyandry come from their parents and the blood .relationships of 4 families have been determined again by us.3. Because the DNA can be obtained from the urine, excrements, formalin-fixed tissues, bans and blood and there is an identical result in the DNA fingerprinting analysis,and therefore the achievement ushered in a vast vistas in the material resource of DNA fingerprinting analysis of ferocious animals and the animals of being in imminent danger.
