摘 要:目的分析我国周期型马来丝虫DNA多态性. 方法收集长爪沙鼠保种的湖北谷城(H)株、四川乐山(S)株和浙江安吉(Z)株马来丝虫成虫,提取基因组DNA,采用RAPD技术分析DNA多态性. 结果得到17个DNA扩增片段,大小为50~1 000 bp.其中a、c为H、S、Z株共有条带, d、e、f、g为H、S株共有条带,b、h、i为H株特有条带.结论 RAPD分析3个地区株马来丝虫在基因结构上既有同源性,又存在差异,表明我国马来丝虫似有种内分化的趋向.Objective To explore whether Brugia malayi from three endemic areas has differentiation or not at gene level. Methods RAPD analysis was used to study the DNA fragments polymorphs of B. malayi from Hubei(H), Sichuan (S) and Zhejiang (Z). Results 17 polymorphic fragments (50-1000 bp) were obtained by amplification of bound genomic DNA from H, S, Z area as tempalate in the polymerase chain reaction. 9 bands (a,b, c, d, e, f, g, h, i ) were from H area, 6 bands (a, c, d, e, f, g ) from S area and 2 bands (a, c) from Z area. The six RAPD fragments (a, c, d, e, f, g ) from S area were similar to that from H area. The twe bands (a, c) from Z area were the same as a part of the bands of H and S area. 1 strong(b) and 2 slight(h, i ) bands from H area can be clearly distinguishable from S area. Conclusion B. malayi from three endemic areas have autoplidy while they exist differences, too.
