摘 要:用甲醛和十二烷基硫酸钠予处理血清白蛋白(BSA)可使银(Ⅰ)和铜(Ⅱ)离子从它们的氨合络离子中与之生成棕色络合物,用FTIR光谱和uv/vis光谱研究了BSA—Ag(Ⅰ)—Cu(Ⅱ)配合物的生成机理,并用分光光度法测定了配合物的形成和还原为金属凝胶的反应性,BSA氨基酸残基上的氮和氧与Ag(Ⅰ)和Cu(Ⅱ)离子形成灵敏度很高的配合物,对氨基的修饰作用证明了这一机理。Pretreatment of bovine serum albumin (BSA) with formaldehyde and sodium lauryl sulphate (SDS) markedly enhances the formation of Ag(Ⅰ)—Cu(Ⅱ) brown complex from their ammoniacal complex. The mechanism for BSA—Ag—Cu complex forming has been developed FTIR spectrum and uv/vis absorbance spectra. And the rate of formation and reactivity of reduction metallic colloidal have been determined by spectrophotometry. It is formed a sensitive complex that Netrigen and Oxygen atoms in amino acid residues of BSA with silver and copper ions. It was proved that the mechanism on modification of amino group. The work will give some theoretioal bases for protein metal staining technologies.
