摘  要:目的探讨G luR-B亚单位表达变化在癫痫形成中的机制。方法采用W estern b lot方法研究G luR-BΔECS.Forebrain小鼠海马G luR-B亚单位的表达变化。结果(1)TgC re4和G luR-Bneo小鼠杂交子代中约25%是G luR-BΔECS.Forebrain小鼠,该鼠系从出生后18~120d内均出现癫痫发作,而且小鼠最终因癫痫频繁发作而导致死亡。(2)W estern b lot半定量结果分析显示,转基因小鼠海马G luR-B亚单位水平较对照组平均下降约25%(P<0.01)。结论G luR-B亚单位Q/R位点编辑障碍和表达降低决定了G luR-BΔECS.Forebrain小鼠癫痫发作的时间、特点和预后。Objective To investigate the rule of GluR-B transcription and expression in seizures of GluR- B^△ECS.Forebrain mice during their development. Methods By employing a semiquantitative immunobloting technique, the expression of GluR-B subunit were determined in hippoeampus of GluR-B^△ECS.Forebrain mice. Results Approximate twenty-five percent offsprings of Tg^cre4 mice crossing with GluR-Ba^△ECS.Forebrain mice were GluR- B^△ECS.Forebrain mice. Seizures oceured hetween P18 and P120 in all GluR-B^△ECS.Forebrain mice, and the high frequencies of seizures finally led them to die off. By taking health littermates as control, approximate twenty-five percent GluR-B subunit was decreased in hippocampus of GluR-B^△ECS.Forebrain mice ( P 〈 0. 01 ). Conclusion Seizures of GluR-B^△ECS.Forebrain mice ware determined by Q/R site uneditable form and low expression of GluR-B subunit in hippocampus.
