摘  要:目的:探讨复方薤白胶囊(CLOC)对野百合碱(Monocrotaline MCYT)诱导的肺动脉高压(PH)大鼠肺组织匀浆中SOD活性、MDA和HYP含量的影响.方法:SD(Sprague-Dawley)大鼠60只,随机分为5组,MCT进行一次性腹腔注射(按60mg/kg),正常对照组一次性腹腔注射等量的生理盐水.于造模第2天开始给药.给药第21天,随机每组取6~8只大鼠进行平均肺动脉压(mPAP)和平均右心室压(mRVP)的测定,测定新鲜肺组织中超氧化物岐化酶(SOD)活性、丙二醛(MDA)和羟脯氨酸(HYP)的含量.另一部分用以观察肺小动脉及右心室病理变化.结果:模型组与正常对照组相比,mPAP明显增高,有非常显著性差异(P<0.01),SOD活力显著下降(P<0.01),模型组小动脉及右心室厚度明显增厚,小动脉管腔甚至闭塞,肺组织炎性细胞浸润较明显;匀浆中MDA、HYP含量增高不明显(P>0.05).CLOC大、小剂量组与模型组相比,mPAP明显下降,有非常显著性差异(P<0.01),匀浆中MDA明显下降,SOD活性显著升高(P<0.01),CLOC大、小剂量组小动脉及右心室厚度明显减轻;肺组织炎性细胞浸润减轻,组间比CLOC大剂量组优于CLOC小剂量组.HYP各组之间无差别(P>0.05).结论:CLOC具有有效的降低MCT诱导大鼠PH的作用,降低肺组织中的MDA含量,增加SOD活性,改善肺小动脉及右心室重构,减轻MCT引起的淋巴细胞浸润现象.可能的机制是CLOC清除自由基、抗脂质过氧化,保护EC,维持VSMC的正常舒张功能.提示CLOC将是防治PH的有效中药.Objective:to explore the effects of Compound longstemen onion capsules (CLOC) on superoxide dismutase (SOD) activity, malondialdehyde (MDA) and hydroxyproline (HYP) concentration in pulmonary homogenate in rats with pulmonary hypertension induced by monocrotaline(MCT). Methods:SD (Sprague- Dawley) rats were randomly divided into five groups: the normal group rats were given normal saline (1 ml/100g/day)the other group were injected in abdominal cavity with a single dose of MCT(60mg/kg), then the rats was treaded with gastric infilsion of normal saline, defferent dosage of CLOC or captopril respectively,22 days after injection of MCT, survey mean pulmonary artery pressure(mPAP) ,mean right ventricle pressure( mRVP), activity of SOD, concentration of MDA and HYP in pulmonary nomogernate were measured by colorimetric analysis ; histopathologic changes in lung, small pulmonary artery and right ventricle was observed. Results :22 days after injection of MCT ,the mPAP was increased significantly elevated compared with normal control group( P 〈 0. 01 ), the activity of SOD was reduced ( P 〈 0. 01 ), the contents of MDA did not changed ( P 〉 0. 05 ), the thickness of small pulmonary trtery and right ventricle observed were thicker in the model group than that in the normal group. The contents of MDA were reduced and the activity of SOD was increased significantly( P 〈0. 01 ), mPAP was decreased significantly elevated compared with model group( P 〈0. 01 ). The thickness of small pulmonary artery and right ventricle observed were thinner in the treaded groups than that in the model group, the contents of HYP were not difference among all groups ( P 〉 0. 05 ). Conclusions CLOC could prevent PH induced by monocrotaline effectively,the activity of SOD was increased, the concentration of MDA was reduced, its mechanisms may be inhibiting lipid peroxidation and clearing oxygen free radical, preventing endothelial cells (EC), maintaining the
