摘  要:以苏丹草幼穗为外植体材料,研究幼穗发育期以及配比的激素组合对愈伤组织诱导发生、生长状态及其绿苗分化能力的影响.结果表明:长度为1.5~5.0 cm的幼穗,在愈伤组织诱导培养基中添加 3.0 mg/L 2,4-D和 0.2 mg/L KT,诱导频率达80%~90%,继代培养基中仍添加 3.0 mg/L 2,4-D+ 0.2 mg/L KT,愈伤组织增殖速度快,且保持植株再生的能力;在愈伤组织分化培养基中添加 2.00 mg/L 6-BA+0.01 mg/L NAA或添加 2.00 mg/L CPPU+0.01 mg/L NAA,经继代培养3~5代的愈伤组织绿苗分化率保持在50%以上;在继代培养中选择外表呈白色、干燥、紧密、颗粒状的愈伤组织,是提高其再生植株能力的有效方法.Immature inflorescences of Sorghum sudanese were used for explants. Effect of development period of immature inflorescences and hormone compositions of different kinds and ration on callus induction, growth state and plant regeneration was studied. Results indicated that the frequency of callus induction reached 80% -90% in the callus induction medium supplemented with 3.0 mg/L 2,4-D and 0. 2 mg/L KT by 1.5-5 cm immature inflorescences. During subculture, rapid growth of callus and plant regeneration were maintained in the callus subculture medium added 3.0 mg/L 2,4-D and 0.2 mg/L KT. The rate of green plant regeneration from callus of the third to fifth callus subcultures was above 50% in the differentiation medium supplemented with 2.00 mg/L 6-BA + 0.01 mg/L NAA or 2. 00 mg/L CPPU + 0.01 mg/L NAA. Selecting the callus of dry, compact, small pellet was a simple effective method in early generation to overcome the obstruction of plant generation.
