摘 要:目的:观察肾康注射液对高糖培养系膜细胞肥大的影响。方法:将体外培养的大鼠肾小球系膜细胞分为空白对照组、甘露醇组、高糖组、高糖+缬沙坦组及高糖+肾康注射液高、中、低剂量组。应用流式细胞术观察细胞大小、周期分布,用[3H]胸腺嘧啶核苷([3H]TdR)及[3H]亮氨酸([3H]Leu)掺入法反映细胞DNA及蛋白质合成情况。结果:高糖组系膜细胞G1期比例增高,G2期、S期比例下降,蛋白质合成增加,DNA合成减少,细胞体积增大;肾康注射液组与高糖组比较,细胞周期分布趋于正常,蛋白质合成减少,DNA合成增加,细胞体积减小。结论:肾康注射液可抑制高糖培养系膜细胞的肥大反应。Objective: To investigate the effects of Shenkang injection on hypertrophy of rat glomerular mesangial cells cultured in high glucose. Methods: Cultured rat glomerular mesangial cells were divided into 7 groups: Blank control group, Mannitol - treated group, High glucose - treated group, High glucose plus Valsartan - treated group, High glucose plus high, middle and low dose of Shenkang injection groups. Every group was treated for 72 hours. The cell volume and proportion of every cell cycle phase were measured by flow cytometry. Incorporation of ^3H - TdR and ^3 H - Leu were determined respectively. Results: High glucose caused proportion of cells in G1 phase increased, proportion of cells in G2 and S phase decreased, incorporation of ^3H -TdR reduced, incorporation of ^3 H - Leu augmented, cellular size enlarged. Shenkang injection may decrease ceils proportion in phase G1, increase that in phase G2 and S, and reduce incorporation of ^3 H - Leu and cellular size, increase incorporation of ^3 H - TdR. Conclusion: Shenkang injection can inhibit hypertrophy of glomerular mesangial cells cultured in high glucose.
