摘 要:目的:动态观察常规方法(冲击量+维持量)补充肌酸对运动训练大鼠自身肌酸生物合成和代谢的影响。方法:将60只大鼠按体重随机分为6组:3组为补充肌酸组,分别以冲击量(1.5g/kg/d)1周、冲击量(1.5g/kg/d)1周+维持量(0.15g/kg/d)4周和冲击量1周+维持量(0.15g/kg/d)8周补充肌酸;其余3组为相应对照组。各组大鼠每天进行游泳训练4小时。分别于实验1周、5周、9周后测定大鼠肾脏精氨酸-甘氨酸脒基转移酶(AGAT)活性,肾脏、肝脏和血清胍乙酸(GA)含量,肾脏、骨骼肌和血清肌酸含量,血清和骨骼肌肌酸激酶(CK)活性及血清肌酐水平。结果:冲击量补充肌酸可导致运动训练大鼠肾脏AGAT活性以及肾脏、肝脏和血清胍乙酸含量降低,随着维持期延长,上述指标逐步恢复至对照组水平。补充肌酸引起肾脏肌酸含量明显增加,肾脏和血清肌酸含量与AGAT酶活性呈明显负相关。结论:冲击量补充肌酸可抑制运动训练大鼠内源性肌酸的合成,而在维持量阶段抑制状态逐渐缓解。建议运动员补充肌酸时不使用冲击剂量而只使用维持剂量。Objective To investigate dynamically the effects of routine creatine (Cr) supplementation with loading + maintaining doses on the endogenous biosynthesis and metabolism of Cr in exercise - training rats. Methods 60 rats were randomly divided into 6 groups according to their weights. Among them 3 groups were differently supplemented with Cr: or loading dose (1.5g/kg/d) for 1 week, or loading dose for 1 week following maintaining dose (0.15g/kg/d) for 4 weeks, or loading dose for 1 week following maintaining dose for 8 weeks. The other 3 groups were treated as the controls without Cr supplementation. All 6 groups of rats were trained by swimming for 4 hours each day during the Cr supplementing period. The rats were respectively killed at the ends of 1, 5 and 9 weeks. L - arginine:glycine amidinotransferase (AGAT) activity in kidney, guanidinoacetic acid (GA) contents in kidney, liver and serum, Cr contents in kidney, skeletal muscle and serum, creatine kinase (CK) activities in skeletal muscle and serum, and creatinine level in serum were determined respectively. Results AGAT activities in kidney and GA contents in kidney, liver and serum were lowered by the loading dose of Cr supplementation in the exercise trained rats, but gradually retuned to the levels in the control rats during the maintaining term of Cr supplement. Cr supplement resulted in increased Cr contents in kidney. The Cr concentrations in kidney and serum had markedly negative correlation with AGAT activities in kidney. Conclusions The endogenous biosynthesis of Cr in the exercise - training rats could be suppressed by Cr supplementation at the loading dose, and be gradually relieved at the maintaining term. It is suggested that athletes should use maintaining dose, for Cr supplementation.
