复方丹参滴丸对胰岛素抵抗犬肝脏保护作用研究

摘　　要：目的通过高脂喂养复制胰岛素抵抗犬模型,观察肝脏损害程度并了解复方丹参滴丸是否在胰岛素抵抗条件下对肝脏具有保护作用。方法选Beagle犬18只,正常对照组(6只)给予普通饲料喂养,12只给予高脂喂养6周,经高胰岛素钳夹实验证实出现胰岛素抵抗后分为:高脂喂养组(6只)继续给予高脂喂养,复方丹参滴丸(6只),在高脂喂养同时给予复方丹参滴丸40粒/天,4个月后处死动物,取肝脏标本进行形态学观察,于第0、6、10月检测生化及氧化指标。结果经高脂喂养后,动物体重、总胆固醇、甘油三酯均明显升高,胰岛素敏感性降低。给予复方丹参滴丸后,动物总胆固醇、甘油三酯显著降低,但体重、胰岛素敏感性与高脂组无显著差异;复方丹参滴丸能有效抑制高脂喂养造成的氧化应激状态。肝脏HE染色结果高脂喂养组出现严重的肝细胞浊肿、肝脏结构破坏以及炎性细胞浸润,复方丹参滴丸对此有明显的改善作用,电镜结果显示,复方丹参滴丸能明显减轻肝细胞脂肪变性。结论本研究表明,复方丹参滴丸在胰岛素抵抗条件下具有明确的肝脏保护作用。机制为复方丹参滴丸通过调脂作用抑制细胞脂肪变性的发生以及通过抗氧化作用减轻细胞损伤程度。Objective To evaluate the protective effect of Cardiotonic Fills against liver steatosis in canine model with insulin resistance induced by a high fat feeding. Methods 12 Beagles were fed with highfat diet for 6 months, insulin resistance was identified by hyperglycemia clamp test. 6 of them received additional cardiotonic Pills for 4 months. Liver samples were obtained for morphological study. Results A panel of metabolic disorders was identified in the high-fat feeding dogs that include obesity, hyperlipidaemia and insulin resistance. Oxidative stress was also shown at the end of 6th months, that is charactered by increase in plasma MDA, and decrease in erythrocyte GSH. The treatment of Cardiotonic Pills significantly decreased the plasma triglyceride and cholesterol level, and reversed the oxidative stress, but with no effect on the insulin sensitivity. In the morphological study, the cardiotonic pills reduced the liver steatosis compared with high-fat feeding control. Conclusion Cardiotonic Pills can protect the liver from steatosis in the current insulin resistance canine model. The lipid-lowering and anti oxidative effects of the Cardiotonic Pills is the underlying reason.
