补肾益气活血方对兔实验性膝骨关节炎软骨细胞bFGF-mRNA表达的影响

摘　　要：目的:探讨补肾益气活血方对膝骨关节炎的软骨细胞中bFGF-mRNA的表达的影响。方法:新西兰兔32只,随机分成四组:假手术组、模型组、醋氨酚组和补肾益气活血方组。其中模型组、醋氨酚组和补肾益气活血方组建立兔Hulth膝骨关节炎模型,分别采取相应的处理方法。术后4周处死取材进行bFGF-mRNA原位杂交,并采用图像分析系统对其表达测定。结果:补肾益气活血方组关节软骨中bFGF-mRNA的阳性表达高于假手术组和模型组(P<0.01),具有显著性差异。结论:补肾益气活血方可上调关节软骨中bFGF-mRNA的表达,从而起到防治膝骨关节炎的作用。Objective： In order to research the influence of bFGF - rnRNA expression in articular cartilage of the rabbit knee osteoarthritic model with Bushenyiqihuoxue recipe treatment. Method： 32 New Zealand rabbits were divided into four groups - sham operated, untreated group, acetaminophen and Bushenyiqihuoxue group at postoperation. All rabbits were established the Hulth knee osteoarthritic model except the sham operated group, The four groups were deal with different therapy respectively and taken samples at postoperation 4 weeks by killing rabbits. All samples were undertaken hybridization in situ and image analysis methods in order to test their bFGF - mRNA expressing intensity. Result： The size of bFGF - mRNA expressing positive degree of Bushenyiqihuoxue group was the highest in all groups （ P 〈 0.01 ）. Conclusion： Bushenyiqihuoxue recipe can prevent and treat the knee osteoarthritis by up- regulating the expression of bFGF - mRNA.
