摘 要:为探讨一氧化氮(NO)在高钙致鸡痛风中的作用,利用已建立的鸡原代肾小管上皮细胞的体外研究模型,经不同浓度的钙进行处理,通过M TT比色法观察了高钙对细胞活性的影响;采用硝酸还原酶法测定了不同浓度高钙处理原代肾小管上皮细胞后培养上清液中亚硝酸根/硝酸根(NO2-/NO3-)的含量(NO的静态氧化形式)。结果表明,分别用4.0 mm o l/L(试验组Ⅰ),8.0 mm o l/L(试验组Ⅱ),12.0 mm o l/L(试验组Ⅲ),16.0 mm o l/L(试验组Ⅳ)C a2+处理细胞,各时段组的细胞活性与对照组相比均显著降低(P<0.01),且细胞外钙浓度越高,细胞活性降低越明显。高钙处理细胞24 h后,各组培养液中NO的含量较对照组升高不显著;48 h后,试验Ⅳ组的NO比对照组显著升高;72 h后,各试验组与对照组相比NO的含量均显著升高。这些结果说明,细胞外高钙能够直接损害肾小管上皮细胞,而NO可能在肾小管上皮细胞的损害中起一定的作用。In order to explore the role of nitricoxide (NO) in avian gout caused by high dietary calcium,the cells culture system of new-born chicken's primary renal tubule was established in vitro,which was used as the model of avian renal tubular cells' injury caused by high extracellular calcium. The viability of the cells was examined by MTT colorimetric analysis,and NOr/NOr ,the stable oxide formation of NO ,in the culture medium was measured with color reaction method(enzyme substant reaction). The results showed that the viability of renal tubular cells,which were exposed to 4.0 mmol/L(experimental group Ⅰ),8. 0 mmol/L(experimental group Ⅱ ),12- 0 mmol/L(experimental group Ⅲ ) and 16.0 mmol/L(experimental group Ⅳ of calcium in medium,were inhibited significantly(P〈0. 01 ). With the increase of extracellular calcium,the level of NO in medium increased. But there was no difference in different groups at 24 h incubation time. In group Ⅳ ,the level of NO in medium were higher than those of control group at 48 h incubation time. The level of NO in experimental group medium was significantly higher than those of control group at 72 h incubation time. It is concluded that high extracellular calcium can inhibit the viability of renal tubular cells ,and NO may play a role in injure of renal tubular cells by high extracellular calcium.
