摘 要:目的:探讨大鼠黑质γ-氨基丁酸神经元的增龄变化规律,为进一步揭示黑质病变的病因提供客观依据。方法:实验于2002-01/2003-07在贵阳医学院解剖学教研室完成。选择健康SD大鼠32只,分为一两个月龄、四五个月龄、十一二个月龄、≥24 个月龄4组,常规石蜡切片,中脑黑质连续冠状切片,进行苏木精-伊红染色、γ-氨基丁酸免疫组织化学染色,图像分析仪测量γ-氨基丁酸免疫阳性产物的吸光度和γ-氨基丁酸免疫反应阳性神经元的胞面积、体密度、数密度、圆球度,并对苏木精-伊红染色细胞进行计数。结果:32只大鼠均进入结果分析。4个月龄组大鼠随年龄增长,中脑黑质神经元数量减少,γ-氨基丁酸免疫反应阳性神经元平均吸光度减少 (P<0,001),γ-氨基丁酸免疫反应阳性神经元数密度、体密度、圆球度、平均体积均减少,平均截面积增加(P<0.01),呈增龄性变化。结论:黑质γ-氨基丁酸神经元随年龄增长而出现的衰老性变化,可能是黑质病变的原因之一。AIM: To study the regularities of GABAergic neurons in substantia nigra of midbrain with age increasing, so as to further provide objective evidence for revealing lesion of substantia nigra. METHODS: The experiment was carried out in the Department of Anatomy, Guiyang Medical College hctween January 2002 and July 2003. Thirty-two healthy SD rats were selected and divided into four groups: 1-2 months, 4-5 months,11-12 months and ≥ 24 months groups. Routine paraffin were made for hematoxylin and eosin (HE) staining and gamma-aminobutyric acid (GABA) immunohistochemical staining, the absorbance of GABA immune positive product and the area, body density, number density, globular degree of the GABA-positive neurons were measured with image analyzer, and the HE stained cells were counted. RESULTS: All the 32 rats were involved in the analysis of results. With the increasing age, the number of neurons in suhstantia nigra of midbrain in rats of 4 months old was decreased, the average absorbance of GABA- positive neurons was reduced (P 〈 0.001), numher density, body density, globular degree, average volume of GABA-positive neurons were all reduced, but the average sectional area was increased (P〈 0.01). CONCLUSION: The increased expression of GABA neurons in substantia nigra with aging may have important implication for the pathogenesis of substantia nigra disease in the region.
