摘  要:目的探讨不同强度训练条件下豚鼠跟腱胶原及蛋白多糖的变化。方法强化训练豚鼠后,切取其双侧跟腱,分别用天狼猩红及甲苯胺蓝染色,偏振光显微镜下观察Ⅰ、Ⅲ型胶原及蛋白多糖的变化,行半定量分析;原子力显微镜观察跟腱超微结构。结果训练后Ⅲ型胶原含量:A组(16.71±1.37)%、B组(13.43±3.16)%与对照组(9.57±1.90)%相比明显增加(P<0.05);高强度训练C组(29.57±6.94)%、D组(18.14±3.19)%增加最多(P<0.01);并且成纤维细胞及黏多糖也增多;高强度下胶原纤维排列紊乱,在相位像上可见带状物质与胶原纤维呈多维编织排列,这种物质是蛋白多糖的侧链。结论强化训练能加速肌腱胶原及蛋白多糖的转化,胶原纤维和蛋白多糖之间存在复杂的编织排列。Objective To investigate the changes of collagen and proteoglycan of Guinea pig' s Achilles tendon suffering from different loads of training, Methods Achilles' tendons of Guinea pigs were harvested after training, stained by picrosirius and toluidine blue respectively. Collagen Ⅰ , Ⅲ and proteoglycan were observed by polarization microscopy and atomic force microscopy (AFM) and analyzed semiquantitatively. Results After enforced training, the relative content of collagen Ⅲ was( 16.71 ± 1.37 ) % in Group A and ( 13.43 ± 3.16) % in Group B, that were significantly increased compared with (9.57 ± 1.90)% in control group ( P 〈 0. 05 ). The most significant increase of the relative content of collagen Ⅲwas seen in Group C and Group D, with ( 29.57 ±6.94)% and ( 18.14 ±3.19)%, respectively( P 〈0. 01 ). In the meantime, fibroblasts and glycosaminoglycans were increased too ( P 〈 0. 01 ). After enforced training, the fibrils become disordered, as was showed by AFM's phase graph that some chains could be seen in and between fibrils, weaving with fibrils multi-dimensionally. Such chains might be side-chains of proteoglycan. Conclusion Enforced training can speed up the convert of collagen and proreoglycan of tendon, where an intricate interweaving exists.
