摘 要:目的:了解高胰岛素对人内皮细胞分泌前列环素(PGI2)和内皮素1(ET1)的影响。方法:采用酶消化法收集人脐静脉内皮细胞,经原代培养后随机分为4组:正常对照组,高胰岛素组,正常+谷胱甘肽组,高胰岛素+谷胱甘肽组;放免法测定4种培养基培养24h,48h,72h时培养基中的PGI2和ET1含量。结果:①高胰岛素组较正常对照组PGI2显著下降(P<0.01),ET1显著升高(P<0.01);②高胰岛素+谷胱甘肽组较高胰岛素组PGI2显著升高(P<0.05)和ET1显著下降(P<0.05)。结论:高胰岛素对血管内皮细胞有损伤作用,还原型谷胱甘肽可减轻这种损伤。Aim: To investigate the influence of high insulin on human umbilical vein endothelial cells(HUVEC) secreting prostacyclin ( PGI2 ) and endothelin-1 ( ET-1 ). Methods: After primary culture, the HUVEC were allocated into four groups:normal control group, normal control with glutathione group, high insulin group, and high insulin with glutathione group; the medium PGI2 and ET-1 in the 4 groups which were cultured for 24 h,48 h, and 72 h were assayed using radioimmunoassay. Results: The content of PGI2 in the high insulin group was lower, and the content of ET-1 was higher com- pared with normal control group (P 〈 0.01 ) ; the content of PGI2 in the high insulin with glutathione group was higher and the content of ET-1 was lower(P 〈 0.05) compared with the high insulin group. Conclusion : High insulin can injure vascular endothelial cells and reduced glutathione can ameliorate the endothelial dysfunction.
