摘 要:目的:观察苯海索体外对人血小板聚集和胞浆游离钙的影响。方法:用比浊度法及钙离子荧光指示剂Fura2AM测定法,分别体外测定苯海索对凝血酶诱导的人血小板聚集及胞浆游离钙浓度的影响。结果:苯海索0.001~1.000mmol·L-1浓度可明显抑制凝血酶诱导的血小板聚集(P<0.05)和胞浆游离钙的升高(P<0.01),均呈剂量依赖性(r=-0.9230,P<0.01;r=-0.9348,P<0.01),其抑制作用可以被钙减弱,而被依他酸增强。苯海索对凝血酶诱导的钙释放无明显影响(P>0.05)。结论:苯海索可抑制人血小板聚集和[Ca2+]i升高,其主要机制可能是抑制细胞外钙离子的内流。Aim : To study the effects of trihexyphenidyl (THP) on aggregation and intracellular free calcium concentration in human platelet. Methods: Using Fura2-AM fluorescence technique and turhidimetric method to examine the effects of THP on aggregation induced by thrombin and intracellular free calcium concentration, respectively. Results: THP(0. 001-1. 000 mmol·L^-1) significantly inhibited the platelet aggregation and the rise of Ca^2+ induced by thrombin in human platelets in vitro in a dose-dependent manner (r = -0. 923 0, P 〈0.01; r = -0. 934 8, P 〈 0.01 ). The inhibitory effects of THP on platelet aggregation induced by thrombin were reduced by adding calcium and increased by adding egtazic acid to the medium, and THP had no effects on intracellular Ca^2+ release induced by thrombin( P 〉 0.05). Conclusion: The inhibitory effects of THP on aggregation and the rise of Ca^2+ in human platelets are mainly due to the inhibition of Ca^2+ influx.
