摘 要:目的:研究发现tamoxifen可延长患者心电图QT间期,易致心律失常,但其离子机制目前还不清楚。本实验将观察tamoxifen对人心房肌细胞超快速激活延迟整流钾电流(IKur)的影响,以探讨tamoxifen影响心肌QT间期的可能机制。方法:通过胶原酶消化法得到单个人心房肌细胞,并通过全细胞电压钳方法记录心肌细胞上的IKur。结果:本法可得到横纹清楚的单个耐钙人心房肌细胞,Tamoxifen(1~30μmol/L)可剂量依赖性并可逆地抑制IKur。结论:Tamoxifen对人心房肌细胞IKur具有显著的抑制作用。AIM: Pervious studies had shown that tamoxifen altered the QT intervals of patients in clinics, but its ionic mechanism is not clear. The present study was to investigate the effects of tamoxifen on ultra-rapid delayed rectifier K^+ curent (IKur) in human atrial myocytes. METHODS: Single human atrial myocytes were obtained enzymetically and IKur was recorded with whole-cell patch-clamp techniques. RESULTS: The single human atrial cardiomyocytes with clear striation were obtained by the present methods, IKur was inhibited by tamoxifen (1 -30 μmol/L) dose-dependently and reversibly. CONCLUSION: Tamoxifen exerts inhibitory effect on IKur in human atrial myocytes.
