摘  要:采用苯酚-硫酸法测定鼠李糖脂生物表面活性剂产生菌发酵液中的总糖含量,研究了显色温度、显色时间、苯酚及硫酸用量对测定的影响.通过试验确定了较佳测定条件:100℃显色15 min,苯酚用量1.0 mL,硫酸用量5.0 mL,于482nm处测定.结果表明,用此方法测定鼠李糖脂生物表面活性剂产生菌发酵液中的总糖含量时,其吸收值与鼠李糖质量浓度(在0~0.04g/L)呈良好的线性关系,相关系数为0.996 9,加样回收率为100.5%(n=5).The content of total saccharides in the fermentation broth for producing biosurfactant rhamnoslipids was determined by phenol - sulfuric acid method. Effects of temperature and time for color development as well as amount of phenol and sulfuric acid to be used were examined. Through experiments, the optimal conditions of determination operation were defined as: temperature and time for color development is 100 ℃ and 15 min respectively and the amount of phenol and sulfuric acid is 1.0 mL and 5.0 mL respectively. The measurement of the absorbance is to be carried at 482 nm. The results showed that the absorbance displays a linear relation against the concentration of rhamnose within range of 0 ~ 0.04 g/L. The correlation co- efficient of standard curve is 0.996 9, and the sample recovery rate achievess 100.5 %(n = 5).
