安胎散提高羊胚胎移植受胎率及其机理的研究

摘　　要：将胚胎移植后的受体山羊60只随机分为2组,试验组和对照组各30只。饲养管理相同条件下,移植胚胎后的第2天将中药“安胎散”拌于精料中饲喂给试验组受体羊,每只羊每天100 g,连用5 d。于两情期后统计其受胎率,试验组为86.7%,对照组为56.7%,差异极显著(P<0.01)。于胚胎移植当日和两情期时采集受体羊外周血样,ELISA方法检测血浆中Th1细胞因子TNF-α、IFN-γ、IL-2的含量,硝酸还原酶法检测NO含量。结果表明,试验组血浆中各细胞因子含量显著低于对照组(P<0.05),NO含量也显著低于对照组(P<0.05);而胚胎移植当日两组羊血浆中细胞因子及NO含量差异不显著(P>0.05)。“安胎散”可显著提高受体羊的受胎率。其机理可能是通过抑制Th1反应,下调Th1细胞因子含量和抑制NO过度合成而实现其安胎作用。After embryo transplanting, 60 goat receipts were randomly divided into two groups, including treated group and control group, with 30 goats in each group. Herbal medicine ＂An Tai San＂ had been fed on the dose of 100 gram per day each for 5 days from the day after the operation. Under the same breeding conditions, the control group wasn＇t fed with Herbal medicine. After two estrous cycles, all animals＇ fecundation rate was registered. The fecundation rate of treated group was 86.7 %, but the controls were 56.7 %. There was significant difference between two groups （P〈0.01）. Their plasma was collected as sample at the time of embryo transplanting for two estrous cycles. The content of cytokines was measured by ELISA and the content of NO was measured through the method of nitric deoxidize enzyme. The content of Th1 cytokines and NO from treated group was significantly lower than the control group after two estrous cycles（P 〈 0.05）. But there was not significant difference between two groups at the time of embryo transplanting（P 〉 0.05）. Our results indicated that the herbal medicine ＂An Tai San＂ has excellent effect on meliorated pregnancy process and promoting the fecundation rate markedly. And the mechanism of herbal medicine ＂An Tai San＂ can inhibit Thl action, decline Thl cytokines content and restrain NO excess produced to benefit normal pregnancy.
