摘 要:目的:①从转录和翻译两个水平研究细胞间黏附分子-1(ICAM-1)、肿瘤坏死因子α(TNF-α)在大鼠肥厚心肌中和在正常大鼠心肌中的表达,并作比较,探讨其表达规律和与心肌肥厚的关系,初步阐述心肌肥厚发生的可能机制。②应用血管紧张素转换酶抑制剂(ACEI)后,重复观察上述指标,探讨ACEI逆转心肌肥厚的可能机制。方法:取SD大鼠30只,雌雄不限,随机分为心肌肥厚组、心肌肥厚+药物组和假手术组3组,每组10只。手术后按要求饲养8周后处死,取心肌标本,行ICAM-1、TNF-α的免疫组化染色;应用RT-PCR法行ICAM-1 mRNA、TNF-αmRNA检测,并进行统计学方法比较。结果:ICAM-1、TNF-α免疫组化结果:心肌肥厚组心肌中2种蛋白表达明显增多,其次是心肌肥厚+药物组;假手术组表达量最少。3组标本ICAM-1mRNA、TNF-αmRNA的RT-PCR检测结果:心肌肥厚组表达量最高,其次是心肌肥厚+药物组,而假手术组表达量最少。结论:ICAM-1、TNF-α参与心肌肥厚的发生和发展。ACEI抑制心肌肥厚的机制与减少ICAM-1、TNF-α的表达有关。Objective:①To study the expression of Intercellular adhesion molecule-1 ( ICAM-1 ) ,Tumor necrosis factor-or(TNF-α) in the hypertrophic myocardium of rat from the level of transcription and translation and the expression of ICAM-1 ,TNF-α in the normal myocardium of rat, and compare them , find the law of expression , inquire into relation between myocardial hypertrophy and ICAM-1 ,TNF-α, and to explain myocardial hypertrophy's pathogenesis preliminarily. ②To observe above items after using ACEI and inquire into pathogenesis of ACEI in reversing myocardial hypertrophy. Methods:30 SD rats were divided into 3 groups and one group contained 10 rats . The first group was myocardial hypertrophic group. The second was myocardial hypertrophic group that was treated with ACEI. The thirth was control. After 8 weeks, they were killed and cardiae specimen were obtained. The proteins of ICAM-1 ,TNF-α were detected by immunohistochemistry and LV's specimen were detected in ICAM-1 mRNA,TNF-α mRNA with RT-PCR. Results: The expression of proteins of ICAM-1 ,TNF-α in the first group in- creased and dropped in the second group. That of the third is the lowest. The expression of ICAM-1 mRNA,TNF-α mRNA was the same to above. Conclusion:ICAM-1 ,TNF-α plays a role in myocardial hypertrophy pathogenesis. ACEI which lowers the expression of ICAM-1 ,TNF-α can results in reversing myocardial hypertrophy.
