摘  要:目的采用高效液相色谱紫外检测法测定人血中瑞芬太尼血药浓度,为临床研究瑞芬太尼提供一种分析方法。方法采用液-液萃取法,用1-氯丁烷提取全血中的瑞芬太尼,选用 GI97559(瑞芬太尼乙基类似物)为内标定量。色谱条件包括色谱柱 Zorba×SB-CN(4.6 mm×250 mm,5 μm),流动相为乙腈-甲醇-磷酸二氢钠缓冲液,柱温30℃,流速1.5 ml/min,紫外波长210 nm 检测,按内标法定量。结果标准曲线在2.5~160 ng/mL 范围内,浓度与峰面积比有良好的线性关系(r=0.998)。最低检测浓度为2 ng/ml。日内差异小于8.6%,日间差异小于9.2%。结论本方法具有操作简便、试剂成本低、灵敏度高、精密度好等优点,为临床研究瑞芬太尼药代-药效动力学提供一种较好的分析技术。Objeetive To establish an HPLC with UV method for determination of remifentanil in human blood. Methods A liquid-liquid extraction method using n-butyl chloride was established. The sample was separated on ZorbaxSBCN (1.6 mm×250 mm,5μn) column with a mobile phase of mixture of acetonitrile, methanol and phosphate buffer, the temperature was set at 30℃ with flow rate of 1.5 ml/min, UV detection at 210 nm. Results The calibration curves were found to be linear at the range 2.5 - 160 ng/ml of remifentnail in whole blood with a good correlation ( r = 0.998).Detection limit was 2 ng/ml. RSD was less than 8.6% within one day and RSD was less than 9.2% at interaction of day. Conclusion This method is rapid, accurate and sensitive, which is very suitable for the investigation of clinical pharmacodynamics of remifentail.
