摘 要:磁黄铁矿因其Fe/S不同,磁性会发生变化.据此,用磁选机将磁黄铁矿按照不同磁感应强度进行分离后,作为选育菌种的磁性培养基,筛选在不同磁性培养基中生长的最适菌群.试验结果表明,强磁性培养基可以选育出以短杆状、无磁性颗粒菌为主的菌群,而弱磁性培养基可以选育出长杆状、有磁性颗粒菌为主的菌群.选育出的菌种与原始菌种相比,完全氧化培养基中亚铁的时间由88 h缩短到30~ 46 h,其中尤以中磁性矿物选育的菌种氧化活性高.磁性培养基在选育菌种的过程中,由于细菌对矿物晶格的浸蚀,磁性会逐渐减弱,因此,要想培养磁性针对性强的菌种,应在驯化过程中采取及时更换磁性颗粒等措施,使培养基磁性保持不变.Pyrrhotite can have change in the magnetism due to its different Fe/S. Based on this, they were separated according to their different magnetization intensities by magnetic separation and used as the magnetic culture media for selecting the strains most suitable to grow in culture media with different magnetism. The test results show that the culture medium with strong magnetism may isolate the strain group with short bar shape and particles with no magnetism as the main while the medium with weak magnetism may isolate the strain group with long bar shape and particles with magnetism. The isolated strains can shorten the time for the full oxidation of Fe^2+ in the culture medium to 30 - 46 hours from 88 hours needed by the original strain. Especially,the isolated strain from the mineral with medium magnetism has a high oxidation activity. In the process of strain isolation, the magnetism of the magnetic culture medium will become weaker gradually due to the bacteria erosion in the mineral crystal lattice. Therefore, to culture the strains appropriate for different magnetism, measures such as changing magnetic particles should be taken in the strain acclimation so as to maintain the magnetism of the culture media.
