摘 要:目的:探讨γ射线对bcl-2基因的影响及意义。方法:将103钯(103Pd)放射性支架和普通支架分别植入犬的肝外胆管,免疫组化方法检测γ射线诱导犬胆管壁凋亡增殖平滑肌细胞中的bc l-2表达,计数凋亡细胞,计算机图像分析系统检测两组胆管管腔面积。结果:bc l-2表达103Pd支架组较普通支架组为弱,胆管出现增殖平滑肌细胞明显凋亡,肝外胆管无明显狭窄;普通支架组未出现增殖平滑肌细胞明显凋亡,肝外胆管有明显狭窄。结论:bc l-2表达与细胞凋亡发生和细胞对辐射敏感性有关;103Pd放射性支架通过降低bc l-2表达,促进胆管壁增殖平滑肌细胞凋亡,从而抑制肝外胆管狭窄。Objective: To study the effect of γ-radiation on tile expression of bcl-2 and its significance. Methods: 103^pd radioactive stent and normal stent were implanted into tile extrahepatic bile ducts of dogs. The expression of bcl- 2 in the proliferated smooth muscle cells of bile duct was determined by immunohistochemical method. The number of apoptotic cells was counted. The luminal area of bile duct was measured by computer image analysis system. Results:The positive expression rate of bcl-2 in 103^Pd radioactive stent group was higher than that in normal stent group. The number of apoptic cells in 103^pd radioactive stent group was greater than that in normal stent group. There was no obvious bile duct stricture in 103^Pd radioactive stent group. Conclusion:The expression of bcl-2 is associated with the apoptosis of smooth muscle cells and the radiation sensitivity of the cells. 103^Pd radioactive stent may facilitate the apoptosis of proliferated smooth muscle cells and thus inhibit the stricture of extrahepattic bile duct by decreasing the expression of bcl-2.
