摘  要:目的:探讨生物电场(EF)对大鼠主动脉血管平滑肌细胞(VSMC)迁移和增殖的影响。方法:体外培养大鼠主动脉VSMC,EF干预72h,显微成像及图象分析技术检测VSMC迁移情况,免疫细胞化学染色方法观察VSMC增殖核抗原表达情况,观察细胞增殖改变。结果:在含有10%胎牛血清(FBS)的培养基中,在150 mV/mm EF作用下,VSMC向阴极迁移明显,向阳极迁移受抑制。150 mV/mm2、50 mV/mm EF干预下,细胞增殖核抗原较无EF干预培养VSMC无明显改变。结论:EF诱导VSMC定向迁移,对VSMC增殖没有明显影响。Objective: To investigate the effects of direct current electric fields on cell migration and proliferation in cultured vascular smooth muscle cells(VSMC). Methods: Cultured VSMC were exposed to physiological electric fields for 72 h in the presence of DMEM +10% foetal bovine serum (FBS). Then the interval photographs of VSMC were taken and analyzed. The expression of proliferating cell nuclear antigen(PCNA) in VSMC was evaluated by immunohistochemistry. Results: In the media containing 10%FBS, VSMC showed an obvious galvanotropic response. After cultured for 72 h, the cells migrated to the negative pole faster than to the positive pole. But after cultured for 24, 48 and 72 h, the levels of PCNA expression were similar among three groups exposed to the different direct current electric fields. Conclusion: Physiological electric fields can induce VSMC migrating cathodally and have not any significantly effects on VSMC proliferation.
