摘  要:目的:初步探索在高原现场进行体外动物细胞培养的可行性。方法:采用成年家兔肝细胞进行体外原代培养,观察细胞形态及培养上清中ALB和AFP水平变化。结果:细胞接种4 h开始贴壁,8 h开始增殖,胞内核数目增多,6 d已生长满瓶;培养上清中ALB及AFP水平均呈现显著升高,其达峰值时间分别为8 d和6 d。结论:体外培养肝细胞生长、增殖及其生物学功能良好,提示在本驻地海拔高度条件下能够进行体外动物细胞培养。Objective: To study the feasibility of cell culture in vitro at high altitude. Methods: The rabbit hepatocytes were cultured at high altitude, and then the changes of cell shape and levels of the biochemical substances in culture supernatants were investigated at the different time. Results: After cultured 4 h, the hepatocytes began to adhibit to the wall of plant board; after 8 h, the hapatocytes began to proliferate and the number of cell nucleus began to increase; after 6 days, the plant hoard had been suffused with growthing hepatocytes. The levels of alpha - fetoprotein(ALB) and albumin(AFP) increased remarkably and reached to the peak within 8 days and 6 days, respectively. Conclusion: The rabbit hepatocytes can be cultured well at high altitude.
