摘　　要：目的探讨瘦素(leptin)与人外周血单核细胞(PBMC)增殖作用的相关性。方法分离并培养PBMC,采用[3H]TdR掺入试验方法,观察瘦素对PBMC增殖作用的影响。结果瘦素可以促PBMC增殖,也可协同PHA或ConA对PBMC的刺激增殖作用,并在一定浓度范围内呈现剂量依赖性。结论瘦素可以促进PBMC增殖。Objective To Discussion the Correlation between leptin and the proliferation of PBMC. Methods PBMC were isolated from hepartinized venous blood of normal donors by density- gradient sedimentation over FicoU - Hypaque and cultured in the 1640 complete medium. Ceils were than cultured in the 1640 complete medium for 56 h in the presence of leptin and PHA or ConA. The proliferative effect was assessed by means of incorporation of [^3H] thymidine. [^3H] TdR was added and cultured for 16 hours. Ceils were then processed in the harvester to measure the incorporation of [^3H] thymidine. Radioactivity was determined by liquid scintillation counter. Results The results suggested that leptin could affect the proliferation of PBMC and it could also enhance the effects of PHA or ConA on the proliferation of PBMC. Conclusion Leptin can affect the proliferation of PBMC.
