摘 要:目的建立浊度法测定两性霉素B及其脂质体效价的方法。方法啤酒酵母为实验菌,加菌量2%~3.3%(V/V),(37±1)℃培养6~8h测定。结果抗生素线性浓度为0.04~0.10μg/ml,一剂量法、二剂量法的平均回收率分别为103.5%(n=5)、106.6%(n=2),日内RSD分别为1.64%、0.38%,日间RSD分别为2.79%(n=8)、0.87%(n=3)。结论本方法灵敏,快速,可作为测定两性霉素B及其脂质体的效价的方法。Objective To establish a turbidimetric method for the content determination of amphotericin B and liposomal amphotericin B. Method The test organism was Saccharomyces cerevisiae and cultured with fresh medium (pH6. 5). Adjusted spore suspension of Saccharornyces cerevisiae inoculated with a 2% -3.3% (V/V) ratio, at (37±1)℃ for 6-8 hours. Results The linear range was 0. 04-0. 10μg/ml. The precision (RSD) of the standard curve assay and two-level assay was 1.64% and 0. 38% for intra-day, 2.79% (n=8) and 0.87% (n=3) for inter-day and the average recoveries were 103.5% (n=5) and 106.6% (n=2) respectively. Conclusion The method was sensitive, rapid and feasible to determine amphotericin B and liposomal amphotericin B
