摘 要:目的通过对静脉和球后2种途径接种白血病细胞建立模型的方法进行比较,探讨建立急性粒单核细胞性(粒单)白血病动物模型的有效方法。方法BALB/c小鼠分别经尾静脉和球后接种1×106WEHI-3细胞,观测其生存天数,光镜监测BALB/c小鼠外周血白细胞计数、未成熟粒单核细胞百分率及骨髓原始粒单核细胞百分率,电镜观测其骨髓白血病细胞形态。结果由静脉途径接种的小鼠生存时间要比由球后途径接种的小鼠明显延长,达到38.40±2.21d;其外周血白细胞计数和未成熟粒单核细胞百分率在第14天明显升高,分别为(19.24±1.73)×109/L和(6.24±0.85)%,并有上升的趋势;其骨髓原始粒单核细胞百分率亦升高明显,达到(41.3±4.90)%.由球后途径接种的小鼠其生存时间为16±2d,外周血白细胞计数、未成熟粒单核细胞百分率及骨髓原始粒单核细胞百分率一直处于正常范围内。结论WEHI-3细胞经静脉途径植入BALB/c小鼠体内,可形成典型的白血病模型,经球后途径接种则不能形成该模型。Objective To explore the effective method to establish mice myelomonocytic leukemia model. Methods BALB/c mice were administrated intravenously and retrobullbarly at doses of 1×10^6 WEHI-3(myelomonocytic leukemia) cells respectively. The survival time of these mice were observed. The count of white blood cells and the percentage of immature granulocyte and monocyte obtained from tail vein were determined on 1 day before transplanting WEHI-3 cells and 7,14,19,24,28 days after transplanting. Myeloblast and monoblast percentage of bone marrow were checked as soon as these mice died. The morphology of WEHI-3 cells of bone marrow were observed by transmission electron microscopy. Results The survival time of the mice administrated intravenously was 38.40±2.21d,the count of white blood cells and the percentage of immature granulocyte and monocyte obtained from tail vein were raised obviously on 14 day after transplantation, reached respectively(19.24±1.73)×10^9/L and (6.24±0.85)%,myeloblast and monoblast percentage of bone marrow reached (41.3±4.90)%. The survival time of the mice administrated retrobulbarly was 16±2d, the count of white blood cells and the percentage of immature granulocyte and monocyte obtained from tail vein did not raise, myeloblast and monoblast percentage of bone marrow was also in normal range. Conclusion Intravenous injection of WEHI-3 cells(1×10^6) into BALB/c mice successfully induced leukemia in these mice 14-28days later, Retrobulbar injection of WEHI-3 cells(1×10^6) into BALB/c mice can not induce leukemia.
