摘  要:目的探讨低剂量辐射(LDR)对急性粒单核细胞性(粒单)白血病小鼠骨髓的影响,为LDR的临床应用提供理论依据.方法将小鼠粒单白血病细胞系WEHI-3尾静脉接种于BALB/c小鼠建立动物模型.接种后第28天对其行LDR,于照射后第1天处死所有小鼠,通过光镜、荧光显微镜、电镜观测骨髓多项指标.结果 BALB/c小鼠在接种WEHI-3细胞后第14~28天能成功诱导移植性粒单白血病的发生.小鼠骨髓涂片的光镜观测显示50mGy,75mGy和120mGy组其原始粒单核细胞百分率降低,与对照组相比P<0.05.小鼠骨髓涂片的荧光显微镜观察显示50mGy,75mGy和120mGy组其骨髓白血病细胞凋亡率升高,且随着时间的延长小鼠骨髓凋亡细胞百分率有降低的趋势,与对照组相比P<0.05.结论 LDR对粒单白血病小鼠有治疗作用.Objective To study the effects of low dose radiation(LDR) on the bone marrow of mice with myelomonocytic leukemia and provide theoretical base of the clinical application of LDR.Methods BALB/c mice were administrated intravenously at doses of 1×10^6 WEHI-3(myelomonocytic leukemia)cells respectively to induced leukemia. Whole body LDR was carried out on 28 day after transplanting WEHI-3 cells. Allmice were executed on 1 day after LDR. Changes and characters of bone marrow were observed by light microscopy, fluorescence microscopy and electron microscopy. Results Intravenous injection of WEHI-3 cells(1×10^6) into BALB/c mice successfully induced leukemia in these mice 14-28 days later. Myeloblast and monoblast percentage of bone marrow in mice irradiated with LDR 50mGy,75gGy and 120mGy was significantly reduced in comparison with those in control mice (P〈0.05). Exposure to LDR 50mGy, 75mGy and 120mGy caused significant enhancement of apoptosis percentage of bone marrow cells in comparison with those in control mice (P〈0.05). Conclusion The results suggest that LDR has curative effect on myelomonocytic leukemia mice.
