摘 要:建立了血清中双氯芬酸钠毛细管电泳高频电导分析法.对电泳介质的种类、浓度以及操作电压和进样量等影响因素进行了优化.实验采用5 mmol/L乳酸-NaOH(pH 3.8)为缓冲溶液,分离电压为22.0 kV,可在7 min内实现对双氯芬酸钠的分离检测.在最佳实验条件下,双氯芬酸钠的线性范围为1.5~220 μg/mL,检出限为0.5 μg/mL,回收率为92.0%~108%.A method based on capillary electrophoresis with high frequency conductivity detection was developed for determination of diclofenac sodium in blood serum. Major factors for separation and determination were studied and the optimal analysis conditions were obtained. In the buffer of 5 mmol/L lactic acid-NaOH (pH 3.8) and separation potential of 22.0 kV, complete separation of diclofenac sodium was achieved. Under the chosen conditions, Diclofenac sodium was eluted within 7 min, the linear range of diclofenac sodium was 1.5×10^2 - 2.2 × 10^2μg/mL, the limit of detection reached 0.5 μg/mL. The method was used for the analysis of diclofenac sodium satisfactorily with a recovery of 92.0% - 108%.
