摘  要:目的:观察左旋咪唑(Levamisole,LMS)对不同实验性变态反应性脑脊髓炎(experimental allergic encephalomyelitis,EAE)模型中枢神经系统中肿瘤坏死因子α(Tumor necrosis factor-α,TNF α)表达的影响。方法:用豚鼠脊髓匀浆(spinal cords homogenate of guinea pig,GPSCH)加完全福氏佐剂(complete Freund's adjuvant,CFA)免疫Wistar大鼠,制作经典GPSCH-EAE模型,应用免疫组化法观察左旋咪唑对其直接激发或间接促发模型中枢神经系统中肿瘤坏死因子α表达的影响。结果:与对照组相比,TNF α的表达于症状高峰期明显升高,以脊髓灰质和小脑白质为最多,表达在小胶质细胞、浸润淋巴细胞以及神经元细胞上,围绕在血管周围成“袖套状”改变以及弥漫性地表达在皮层下和室管膜下;各实验组之间TNF α的表达量之差异没有显著性(P>0.05)。结论:TNFObjective: To observe the effects of levamisole on the expressoin of tumor necrosis factor-α (TNF α ) in central nervous systerm of different animal model of experimental allergic encephalomyelitis in Wistar rat. Method: Use the spinal cords homogenate of guinea pig (GPSCH) mixed with complete Freund's adjuvant(CFA) immune the Wistar rat to make typical GPSCHEAE model, observe the effects of levamisole on the expression of TNF-α in central nervous systerm was detected by immunohistochemical staining with monoclonal antibody to TNF-α according to specification in the animal model by its direct stimulating or promoting. Result: Compared to the control group, the expression of TNF-α was obviously higher at the peak of symptom, expressed on microglia, infiltrating lymphocyte and neuron, especially in spinal cord and the white matter of cerebellum, forming perivascular cuffing alteration as well as the positive cells under ependyma and cortex.The expression of TNF-α in each experimental group was not obviously significance (F〉 0.05), have concordance. Conclusion: TNF-α had an important role in the morbility process of different EAE.
