摘 要:目的探讨线粒体酶在铝致大鼠凋亡神经细胞中的改变。方法健康雄性SD大鼠40只,按体重随机分为4组:生理盐水、Al3+2.5、51、0 mg/kg组。腹腔注射染毒60 d后处死,用TUNEL法检测细胞凋亡,电镜观察线粒体结构的改变,用试剂盒测定线粒体超氧化物歧化酶(SOD),琥珀酸脱氢酶(SDH),三磷酸腺苷(ATP)酶(总ATP、Na+-K+ATP,Ca2+ATP和Mg2+ATP酶)活力。结果随染铝剂量的增高,凋亡指数(AI)逐渐升高(P<0.05),Na+-K+ATP酶、Ca2+ATP酶、Mg2+ATP酶和SOD均降低(P<0.05);海马神经细胞SDH均数降低,但差异无显著性(P>0.05)。海马神经细胞凋亡指数与Na+-K+ATP酶、Ca2+ATP酶、Mg2+ATP酶和SOD呈负相关(r=-0.439,-0.501,-0.530,-0.815,P<0.05),与SDH没有相关性(r=0.287,P>0.05)。结论铝可以诱导大鼠海马神经细胞凋亡,并引起线粒体结构和线粒体酶活力的改变。Objective To study the changes of mitochondfial enzymes of neurocyte in rats' hippocampus induced by aluminum (Al) in vivo.Methods fourty SD rats,adults and healthy,were divided into 4 groups at random:normal saline,Al^3+ 2.5,5,10 mg/kg. They were exposed to AlCl3 by intrapefitioneal injection, 60 d. After exposure, the apoptosis of neurocyte in rats' hippocampus was measured by method of TUNEL. The structure of mitochondria was observed by electron microscope, superoxidase dismutase ( SOD ), succinate dehydrogenase (SDH), Na^+ -K^+ ATPase, Ca^2+ ATPase and Mg^2+ ATPase were detected by test Kits. Result With the increasing oft he dose of Al, the apoptosis index (AI) of neurocyte in rats' hippocampus increased (P 〈 0.05). The structure of mitochondfia was changed significantly. The levels of Na^+ -K^+ ATPase, Ca^2+ ATPase, Mg^2+ ATPase and SOD decreased (P 〈 0.05). The level of SDH did not significantly decreased ( P 〉 0.05 ). There were negatively correlation between AI and the levels of Na^+ -K^+ ATPase,Ca^2+ ATPase, Mg^2+ ATPase and SOD ( r = - 0.439, - 0.501, - 0.530, - 0.815, P 〈 0.05);but there was no correlation between AI and the level of SDH ( r = 0.287, P 〉 0,05). ConcIusion AI can induce apoptosis of neurocyte in rats' hippocampus, and it can result in changes of the structure of mitochondfia and mitochondfial enzymes.
