摘 要:目的研究丙烯酰胺(ACR)亚慢性染毒对小脑神经丝(NF)蛋白的影响,探讨其中毒性神经病机制。方法以204、0 mg/kg ACR腹腔注射雄性Wistar大鼠2个月,Western Blot法测定小脑中NF-L、NF-M和NF-H相对含量。结果与对照组相比,高剂量组小脑上清液NF-L、NF-M、NF-H和沉淀NF-L分别增高348%、241%、32%和20%(P<0.01),而沉淀NF-H降低14%(P<0.05);低剂量组沉淀NF-L、上清NF-H分别增加16%、17%(P<0.05);低剂量组上清NF-L、NF-M及沉淀NF-M、NF-H均略为降低,但差异无显著性(P>0.05)。结论ACR亚慢性中毒可引起小脑组织中NF亚单位蛋白的明显改变,尤其以上清中NF-L、NF-M升幅最大(348%、241%)。ACR引起的中毒性神经病与小脑组织中NF亚单位的含量变化有关。Objective To explore the neurotoxic mechanism of acrylamide monomer in rats. Methods Male Wistar rats weighing 180 - 230g were randomly divided into three groups, each 15 animals. The rats in group 1 served as control, and received normal saline. The animals in group 2 and 3 were given acrylamide dissolved in physiological saline(20,40mg/kg, ip, 3days/week )for 8weeks. Special tests included the observations of spontaneous locomotion in an open field and the levels of neurofilament subunit proteins were assayed by western blot. Results In 40 mg/kg dosage group, compared to the control group, the levels of neurofilament light(NF-L), NF- M,NF-H in supernatant of rat cerebellum and NF-L in pellet were increased by 348 %, 241%, 32 %, 20 % respectively ( P 〈 0.01 ), whereas the levels of NF-M in the pellet was decreased by 14% ( P 〈 0.05). In 20 mg/kg dosage group, acrylamide monomer produced a 16% increase ( P 〈 0.05) and 17% increase ( P 〈 0.05) in NF-L of the pellet and NF-H of the supernatant respectively. Both NF-L and NF-M in the supernatant, NF-M and NF-H in the pellet lessened slightly, but there was no significant alteration when compared to the control group (P 〉 0.05). Conclusion Acrylamide can alter the levels of NF subunit proteins in cerebellum, which suggests the possible involvement of them in the neuropathy induced by acrylamide.
