β-榄香烯诱导人胃癌BAC823细胞凋亡的实验研究

摘　　要：目的探讨β-榄香烯对人胃癌BAC823细胞的增殖抑制作用及其对细胞凋亡的影响。方法每批细胞按空白 对照组和药物处理组随机分组,其中药物处理组依药物浓度的不同再分为0．01、0．02、0．04、0．08、0．10、0．16 mg·mL-1处理组,作用时间为24 h。采用MTT法检测β-榄香烯对癌细胞的增殖抑制作用;采用流式细胞光度分 析术(FCM)检测细胞凋亡率及细胞周期时相分布,同时进行细胞的形态学观察。结果①MTT的检测结果显示 药物浓度和细胞增殖抑制率间具有正相直线相关关系(P<0．01),β-榄香烯体外作用24 h,对胃癌细胞的IC50为 0．078 mg·mL-1。②FCM检测及细胞形态学观察结果显示β-榄香烯可阻滞BAC823细胞于S期并诱导细胞凋亡, 但细胞凋亡率并不完全与药物浓度成正相关。作用时间为24 h时其诱导人胃癌BAC823细胞发生凋亡的最佳浓度是在 0．04～0．08 mg·mL-1。结论β-榄香烯对肿瘤细胞具有较强的增殖抑制作用,在一定药物浓度范围内,随药物浓度 的增加β-榄香烯可使细胞凋亡率升高,但超过一定作用浓度,药物的细胞毒作用增强,细胞凋亡率则呈下降趋势。OBJECTIVE To explore the effect of β-elemene on the inhibition of proliferation and apoptosis of human gastric cancer BAC823 cells. METItODS The cells were divided into two groups： the blank control group and the drug group. The drug group was divided into 0. 01, 0. 02, 0. 04, 0.08, 0.10 and 0.16 mg · mL^-1 groups. The duration was 24 h. Inhibition of proliferation was measured with MTT assay. Apoptotic rate and fraction of the cell cycle phase were measured with flow cytometry. At the same time, morphological and adhesive changes of the cells were observed under an inverted microscope. RESULTS ①There was positive correlation between the drug concentration and inhibition of proliferation （P〈0. 01）. From the linear regression equation IC50 of BAC823 cells was 0. 078 mg · mL^-1 after in cubation with β-element for 24 h. ②β-elemene could inhibit BAC823 cells at S phase and induce the apoptosis.There was no correlation between the rate of apoptosis and the drug concentration. CONCLUSIONS β-elemene can inhibit the proliferation of cancer cells. Apoptotic rate rises with the increase of drug concentration at certain range. Beyond certain concentration the apoptotic rate decends, necrosis increases.
