摘 要:研究含非甲基化CpG基序的寡核苷酸(CpG-ODN)对慢性乙型肝炎患者(CHB)外周血树突状细胞(den- dritic cell,DC)表型和功能的影响。以CpG-ODN刺激CHB和健康者外周血单核细胞衍生的DC成熟,结果显示,与 non-ODN和PBS组比,CpG-ODN能明显提高CHB患者DC的HLA-DR、CD86表达,使其IL-12分泌增加,刺激T 细胞增殖的能力亦显著增强(P<0.05),但不能明显提高CD1a表达。因此,CpG-ODN与TNF-α一样能促进CHB 患者外周血DC成熟进瓶增强其抗原提呈功能;CHB患者的細胞因子环境可能是DC功能沉默的重要原因。To investigate the effect of immunostimulatory CpG - ODN on the phenotype and function of dendritic cells (DC) in patients with chronic hepatitis B (CHB). CpG- ODN and other control reagents such as TNF-α and non - ODN were added respectively to the culture medium of monocyte - derived DC in CHB and heahhy volunteers. Compared with control groups of non - ODN and PBS, the expression rates of HLA - DR and CD86 of DC in CHB with the treatment of CpG- ODN were obviously in- creased, the IL- 12 level was 'also raised, and the stimulating capacity of DC to allogenic T lymphocytes in mixed lymphocyte reaction(MLR) was improved significantly ( P 〈 0. 05) ; In conclusion, CpG - ODN, like TNF - a, has remarkable stimulatory effect on the phenotype and function of monocyte- derived DC in patients with CHB. The disorder of expression levels of plasma TGF - β and IFN -γ in CHB might be responsible for the functional silence of DC.
