摘 要:目的探讨老龄大鼠基底前脑-皮质胆碱能系统TRKB蛋白表达的老龄性变化,同时对比观察人参皂甙对其改变的影响。方法选用WISTAR♀大鼠30只,随机分为3组(N=10):青年组、老龄组、给药组(第17个月始饲以人参皂甙至25-27月龄)。采用WESTERN BLOT技术结合图像分析对基底前脑、海马结构和大脑皮质进行定量研究。结果老龄组基底前脑、海马结构和大脑皮质TRKB蛋白表达分别较青年组下调了61%、99.7%和57.7% (P<0.01),但给药组上述3个脑区的TRKB蛋白表达较老龄组有所回升,分别较老龄组上调了65%、78.5%和 22.6%(P<0.01)。结论老龄组基底前脑-皮质胆碱能系统TRKB蛋白表达较青年组下调,而人参皂甙可明显上调该3个脑区TRKB蛋白的表达。Objective To investigate the age-associated alterations of TrkB protein in basal-cortex cholinergic system and the effect of ginsenosides (GS) on these alterations. Method Thirty female Wistar rats were randomly divided into 3 groups, young, aged and GS groups(feed GS in drinking water from 17 to 25-27 months old) , each group having 10 rats. Western blotting ad imaging analysis system were used to detect the expression of TrkB protein in baso-forebrain, hippocampal formation, and cortex quantitatively. Results TrkB protein was decreased by 65% ,78.5% ,22.6% (P 〈 0.01 )in baso-forebrain, hippocampal formation, cerebral cortex in aged group compared with that in young group. TrkB protein level was increased by 65% ,78.5% and 22.6% (P 〈 0.01 )in GS group compared with aged group. Conclusion TrkB protein expression was decreased in basal-cortex cholinergic system as compared with young group, which can be upregulated by GS.
