芩连合剂对幽门螺杆菌相关性慢性胃溃疡治疗作用的机理研究

摘　　要：目的:探讨芩连合剂对幽门螺杆菌延缓乙酸性胃溃疡愈合的治疗作用及其作用机制。方法:采用幽门螺杆菌(Hp)多次重复感染乙酸性胃溃疡大鼠,造成Hp相关的慢性胃溃疡模型,运用免疫组化和原位杂交等技术观察该方对Hp感染的慢性胃溃疡大鼠胃黏膜碱性成纤维生长因子(bFGF)mRNA和胃黏膜细胞增殖核抗原(PCNA)的影响。结果:中药组与Hp+乙酸组相比溃疡底部及周围胞浆中bFGFmRNA表达明显增强,并能显著提高胃黏膜PCNA标记指数的阳性率。结论:芩连合剂能有效拮抗NCTC11637株Hp对慢性胃溃疡的延缓愈合,其加速溃疡愈合的主要机理与促进溃疡周围bFGF表达和促进胃黏膜细胞增殖等有关。Objective：To investigate the therapeutic effect of Chinese medicine“QinLianHeJi” （QLHJ）on chronic gastric ulcer healing in rats, mainly focus on the studies on its treament mechanisms. Methods ： A rat chronic gastric ulcer model was made by infecting Hp（NCTC11637 strain）after injecting acetic acid into the subserosa of the stomach. In the experiments, the techniques of immunohistochemical stain and in situ hybridization etc. were used to observe the expression of bFGFmRNA and PCNA in th ulcer. Results ： Comparing each treating group with Hp ading acetic acid control group, the expression level of bFGFmRNA was increased, and they all have a remarkable effect on increasing PCNA. Conclusion ： QLHJ could accelerate the ulcer healing up, and its main mechanism involve the factors by promoting the expression of bFGFmRNA and increasing the cell proliferation of gastric mucosa.
