摘  要:【目的】探讨核仁素反义寡核苷酸对RAW264.7细胞增殖与凋亡的影响。【方法】采用反义寡核苷酸技术以抑制RAW264.7细胞中核仁素的表达后,用MTT法检测细胞增殖状况,流式细胞术及DNA琼脂糖凝胶电泳检测细胞凋亡。【结果】免疫印迹结果显示,反义寡核苷酸导入细胞后24 h,核仁素的表达被显著抑制,同时反义寡核苷酸处理组细胞的增殖能力明显受到抑制,细胞凋亡百分率显著升高,并能检测到清晰的“梯状条带”。而正义及随机寡核苷酸导入细胞后不能降低核仁素的表达,对细胞增殖及凋亡均无明显影响。【结论】核仁素表达下调能抑制RAW264.7细胞增殖并能触发RAW264.7细胞凋亡。[Objective]To clarify the effect of nucleolin antisense oligonucleotides on proliferation and apoptosis in RAW 264. 7 cells. [Methods]After inhibiting the expression of nucleolin by using antisense oligonucleotides, MTT, flow cytometry (FCM) assays and DNA ladder assays were performed to assess the cellular proliferation and apoptosis respectively. [Results]After transfecting with antisense oligonucleotides for 24 hours, immunoblotting showed that the expression of nucleolin was repressed significantly. Moreover, in cells treated with antisense oligonucleotides, the cellular proliferation was remarkably inhibited, apoptotic cell increased significantly and “DNA ladder” could be observed clearly. But sense and random oligonucleotides had no effect on cellular proliferation and apoptosis. [Conclusion]Down-regulation of nucleolin can inhibit cellular proliferation and initiate apoptosis in RAW264.7 cells.
