黄芪对腹膜透析相关腹膜间皮细胞VEGF分泌和表达的影响

摘　　要：【目的】了解黄芪注射液对体外培养的人腹膜间皮细胞血管内皮细胞生长因子(VEGF)分泌及其mRNA表达的影响。【方法】取择期手术患者的网膜间皮细胞体外培养,第三代细胞用于实验。细胞同步后分五组观察,对照组为加入等量的RPMI1640与完全培养基,PDS组加入等量的PDS与完全培养基;黄芪1组加入等量的含黄芪注射液(10 mg/ml)的PDS与完全培养基;黄芪2组加入等量的含黄芪注射液(20 mg/ml)的PDS与完全培养基;黄芪3组加入等量的含黄芪注射液(40 mg/ml)的PDS与完全培养基。细胞培养24 h后,用ELISA的方法检测细胞上清液中VEGF的含量并比较。用逆转录多聚酶链式反应(RT-PCR)的方法检测VEGF mRNA表达情况并比较。【结果】VEGF在对照组的浓度显著低于PDS组[(39.40±11.34)VS(86.64±21.64)ng/mg(P<0.05)]。腹膜间皮细胞经含不同浓度黄芪注射液的PDS干预后,VEGF的分泌与PDS组比较有显著下降,在黄芪1、2、3组分别为(63.98±13.51)ng/mg,(49.76±13.51)ng/mg和(42.38±18.05)ng/mg(均P<0.05)。黄芪2组和3组显著低于黄芪1组(P<0.05)。黄芪3组和2组比较呈下降趋势但无显著差异(P>0.05)。VEGFmRNA表达在PDS组最高,比对照组上升(42.33±11.27)%,显著高于黄芪1,2,3组的(23.81±6.77)%,(14.67±4.32)%和(10.69±5.90)%(P<0.05)。黄芪2组和3组显著低于黄芪1组(P<0.05)。黄芪3组和2组比较呈下降趋势但无显著差异(P>0.05)。【结论】黄芪注射液具有抑制PDS导致的腹膜间皮VEGF过度分泌和表达的作用。[Objective] To investigate the effect of Astragalus on the secretion and mRNA expression of VEGF in cultured human peritoneal mesothelial cells. [Methods]Human peritoneal mesothelial cells （HPMC） got from patients undergone surgical operation were cultured in vitro according to the method introduced by Stylianou. The cells were cultured in RPMI1640 medium containing 20% fetal bovine serum. Ceils from the third passage were used for the experiments, and incubated with RPMI1640 culture medium containing 0. 1% FBS for 24 hours. Then,they were divided into five groups （RPMI1640 culture medium as control, PDS containing 1.5 % glucose as PDS group, PDS containing 1.5% glucose and 10mg/ml Astragalus as Astragalus group 1, PDS containing 1.5% glucose and 20 mg/ml Astragalus as Astragalus group 2 and PDS containing 1.5% glucose and 40mg/ml Astragalus as Astragalus group 3）for testing. Each group was supplemented with equal volume of RPMI 1640 culture medium containing 20% FBS. After 24 hours incubation, the levels of VEGF in supernatant of the cell culture and the mRNA expression of VEGF were detected. [Results]The levels of VEGF in supernatant of the cell culture were significant lower in control group than those in PDS group[（39. 40± 11.34） VS （86.64± 21.64） ng/mg,（ P 〈0. 05）]. Marked lower levels of VEGF were also found in Astragalus groups as compared with the PDS group respectivly, P 〈0. 05. The levels of VEGF in Astragalus group 1 were significant higher than those in Astragalus group 2 and Astragalus group 3（ P 〈0. 05）. The level of VEGF in Astragalus group 3 was lower than that in Astragalus group 2, but no significant difference was found between them（ P 〉0. 05）. Higher mRNA expressions of VEGF were found in PDS group compared with the group of control and Astragalus groups. In PDS group , the mRNA expression of VEGF increased significantly to 132.33 % as compared with the control group.
