摘  要:建立了反相高效液相色谱法(HPLC)同时测定红直獐牙菜中的獐牙菜苦苷、龙胆苦苷、芒果苷含量的方法.在检测波长254 nm、柱温30℃、流速1 mL/min时,用ZORBAX SB-C18(250mm×4.6mm i.d.,5μm)柱,以MeOH和H2O(含0.04%H3PO4)在0~20 min内由 20%至35%线性梯度洗脱.上述3种成分均达到基线分离,獐牙菜苦苷、龙胆苦苷、芒果苷的线性范围分别为0.0095~2.9μg(r=0.9999),0.0486~2.56μg(r=0.9999),0.0056~2.8μg(r=0.9999);回收率为97.7%(RSD=4.3%),99.5%(RSD=3.5%),103%(RSD=1.1%).To establish a quantitative method of simultaneously determination of swertiamarin, gentiopicroside and mangiferin in Swert/a erythroticta Maxim by RP - HPLC. The samples were separated on the column of Zorbax SB- C18 (250 mm × 4.6 mm i.d., 5μm) which were diluted with methanol and water (0.04% phosphoric acid).The ratio of methanol and water increased from 20:80 to 35:65 in 20 min with a detected wavelength of 254 nm; flow rate of 1 mL·min^-1 ;column temperature of 30℃. Three compounds were base - isolated. The linear ranges of swertiamarin, gentiopicroside and mangiferin were 0.0095 - 2.9 μg(r = 0.9999) ,0.0486 - 2.56 μg(r = 0.9999) ,0.0056 - 2.8 μg(r = 0.9999) respec- tively;The average recoveries were 97.7% (RSD = 4.3%), 99.5% (RSD = 3.5%), 103% (RSD = 1.1% ) respectively.
