摘 要:腹腔注射10~40mg.kg-1.d-1半叶马尾藻多糖[Sargassum hemiphyllum(Turner)C.Agardh poly-saccharides,SHP],7d后,小鼠经5Gyγ射线照射,观察SHP对辐射损伤小鼠骨髓CD3+4细胞占骨髓有核细胞的百分率、CD34基因表达和SCF(stem cell factor,干细胞因子)mRNA基因表达的变化,探讨SHP抗辐射损伤保护小鼠造血功能的机理。用流式细胞术测定CD34+细胞含量,通过RT-PCR检测CD3+4和SCF mRNA基因表达。结果显示SHP能显著提高辐射小鼠CD3+4细胞的百分率,提高CD3+4和SCF mRNA基因的表达。增加CD3+4细胞数、促进CD3+4和SCF mRNA基因表达可能是该海藻多糖抗辐射、保护小鼠造血功能的重要机理之一。The mice were administered with 10 - 40mg·kg^-1·d^-1 SHP [Sargassum hemiphyllum (Turner) C. Agardh polysaccharides, SHP] ip for 7d before 5Gy γ-rays irradiation to observe the effect of SHP on the mRNA expression of CD34^+ and SCF(stem cell factor), and to study the mechanism of SHP against irradiation on the hematopoietic system in irradiated mice. Flow cytometry was used to detect number of bone marrow cells and the mRNA expression of CD34^+ and SCF were analysed by RT-PCR test. The results showed that the percentage of CD34^+ cells was decreased by γ-rays irradiation(5Gy) and SHP(10-40mg·kg^-1) could increase significantly the percentage of CD34^+ cells to exhibit a protective effect of SHP on bone marrow in irradiated mice. Expressions of CD34^+ and SCF mRNA were lower in model group than those in conrol group and the expressions were higher in three test groups than those in model group. The results indicated that the increases of expressions of CD34^+ and SCF mRNA could be a mechanism of SHP against irradiation on the hematopoietic system in mice.
