摘 要:目的探讨高能冲击波(HESW)致肾细胞凋亡与氧化应激损伤的相关性。方法制成单肾的40只家兔随机等分为对照组、1 000次与2 000次HESW组和2 000次HESW+别嘌呤醇预处理组。HESW冲击后24 h观察受冲击肾组织超氧化物歧化酶(SOD)活性和脂质过氧化终产物丙二醛(MDA)含量,原位缺口末端标记法(TUNEL)和流式细胞术检测凋亡细胞。结果与对照组比较,HESW组肾组织MDA和细胞凋亡率增加、SOD活性降低(P<0.01),2 000次HESW组上述变化更明显,别嘌呤醇预处理组上述变化明显改善(P<0.05,P<0.01)。肾MDA含量与细胞凋亡率呈正相关关系(r=0.79,P<0.05)。结论HESW所致的肾细胞凋亡与氧化应激损伤有关;HESW次数增多肾细胞凋亡增加,抗氧化剂预处理可减少凋亡。Objective To investigate the relationship between oxidative stress and cell apoptosis in the kidney of rabbit induced by high energy shock wave (HESW), Methods Forty healthy male rabbits were surgically managed to the mono - nephron models and randomized into 4 groups consisting of ten each : control group, 1 000 HESW group, 2 000 HESW group and allopurinol pretreated plus 2 000 HESW group, The rabbit kidney tissues were obtained, The activity of superoxide dismutase (SOD) and malondialdehyde (MDA) levels in the renal tissue were measured 24 hours 'after HESW application. The cell apoptosis was detected by TdT - mediated dUTP nick end labelling (TUNEL) and flow cytometty. Results Compared with the control group, the MDA level and the rate of apoptosis in the HESW groups increased, and the activity of SOD reduced significantly ( P 〈 0, 01 ). The changes mentioned above improved obviously in the allopurinol pretreated plus 2 000 HESW group compared with those in the 2 000 HESW group ( all P 〈 0. 05 ), There was significantly positive correlation between the apoptotic rates and the MDA levels (r = 0. 79, P 〈 0. 05 ), Conclusion Oxidative stress participates in the process of renal cellular apoptosis induced by HESW. With the increase of the HESW frequency, the cell apoptosis in the kidney of rabbit increases. The antioxidant improves the state of oxidative stress and antagonizes cellular apoptosis in the HESW - treated kidney tissue.
