摘 要:对辐照前、后两种庆大霉素进行体外抑菌实验,研究辐照对庆大霉素效价的影响。应用辐射交联技术, 分别于辐照前、后加入药物,制备含药水凝胶膜,研究其药物体外释放规律。结果表明,37.5 kGy电子束对 庆大霉素的效价无影响。辐照前、后掺加药物的水凝胶膜分别在12h、8 h内药物快速释放达高峰,而后缓慢 释放,持续到第5天,其释放规律符合Higuchi方程,释药原理属骨架型扩散释放机制。相比较而言,辐照前 掺入药物的水凝胶膜药物释放更为完全。Antimicrobial efficacy of irradiated gentimicin was compared with the control and effect of the irradiation on potency of the gentamicin was studied. Hydrogel films containing gentamicin were prepared by radiation crosslinking technique, and mechanisms of the drug release in vitro were studied. The results indicated that potency of the gentamicin irradiated to 37.5 kGy did not change. The hydrogel films incorporated gentamicin before the irradiation released gentamicin to peak during 12 h, while the hydrogel films incorporated gentamicin after irradiation released gentamicin to peak during 8 h. The drug released slowly up to 5 d. The drug delivery conformed to Higuchi kinetic equation, and mechanisms of the drug release were matrix diffusion. By comparison, the hydrogel films incorporated gentamicin before irradiation released the drug more completely.
