摘  要:目的观察低相对分子质量肝素(LMWH)对内毒素(LPS)作用下人脐静脉内皮细胞(HUVEC)表达血管性血友病因子相关抗原(vWF:Ag)、纤溶酶原激活物抑制剂-1(PAI-1)及蛋白 S(PS)的影响。方法常规从脐带分离及培养 HUVEC,取第1~3代 HUVEC 于24孔板进行分组实验。应用酶联免疫吸附法(ELISA)或免疫比浊法测定 HUVEC 上清液中的 vWF:Ag、PAI-1及 PS。结果(1)在 LPS 刺激下,HUVEC 培养上清液中 vWF:Ag 量较对照组增加(由2.0% 增至3.5%),差异有显著性(P<0.05)。不同浓度 LMWH(20 u/ml、50 u/ml、100 u/ml)均可抑制 LPS 对 HUVEC 表达 vWF:Ag 的影响,与内毒素组比较,差异有显著性(P<0.05);(2)在 LPS 刺激下,HUVEC 表达 PAI-1的量增加,与对照组比较,差异有显著性(P<0.05)。加入不同浓度的 LMWH,可下调 LPS 对 HUVEC 表达 PAI-1的影响。各剂量组与 LPS 组比较,差异均有显著性(P<0.05);(3)以1μg/ml LPS 刺激 HUVEC,PS 表达量下降,与对照组比较,差异有显著性(P <0.05)。同时加入 LMWH 可上调 LPS 对 HUVEC 表达 PS 的影响,各剂量组与 LPS 组比较,差异有显著性(P<0.05)。结论 LPS 可上调 HUVEC 表达 vWF:Ag 及 PAI-1,下调 HUVEC 表达 PS。LMWH 能拮抗上述作用。提示:LMWH 对 LPS 诱导的内皮细胞的活化与损伤有保护作用,在重症炎症状态下使用 LMWH 可能有利于器官功能的恢复和预防 DIC。Objective To evaluate the effect of low molecular weight heparin (LMWH) on vitro lipopolysaccharide (LPS) -induced expressions of yon Willeband factor antigen ( vWF: Ag), plasminogen activator inhibitor-1( PAI-1 ) and protein S (PS) on human umbilical vein endothelial cell(HUVEC). Methods HUVEC cells in their first to third passage were incubated with LPS in serum free medium with or without LM- WH. After incubate for indicated time, the supernatant of the cultured cells were collected, the level of vWF: Ag,PAI-1 and PS were measured by ELISA or Immunoturbidimetry. Results ( 1 ) Compared with control group,co-incubate HUVEC with LPS induced the volume of vWF: Ag increasement from 2% to 3.5% (P 〈 0.05) , and compared with LPS group, addition of LMWH at concentrations of 20 u/ml,50 u/ml and 100 u/ml respectively could down-regulate the expression of vWF: Ag induced by LPS ( P 〈 0.05 ) ; (2) Compared with control group,the LPS-induced expression of PAI-1 were up-regulated ( P 〈 0.05 ) , and compared with LPS group, the some doses of LMWH could counteract the up-regulation effect of LPS (P 〈 0.05) ; ( 3 ) Compared with controtgoup the LPS-induced expression of PS were down-regulated(P 〈0.05 ) ,and compared with LPS goup, the same doses of LMWH could up-regulate the volume of PS ( P 〈 0.05 ). Conclusion LPS could upregulate the expressions of vWF : Ag and PAI-1 , but down-regulate the expression of PS. These effects could be counteracted by LMWH. It may shown that LMWH could protect endothelial cells by counteracting the effect of LPS-induced expressions of vWF:Ag,PAI-1 and PS,which may favor the use of LMWH under severe inflammation.
