摘 要:目的研究体外培养大鼠海马神经元在发育过程中,GABA-A受体离子通道的电生理学特性的变化规律。方法用膜片钳记录仪在体外培养的不同时期,记录大鼠海马神经元GABA-A受体离子通道全细胞通道电流、电导的变化。结果海马神经元GABA-A受体全细胞离子通道平均电流,在培养的第3天、第7天、第10天分别为12.04±3.12、18.21±3.68和20.15±4.03 pA,相对于第3天,第7天和第10天分别增加了50%和70%。平均电流从10%到90%的上升时间为2.93±0.23 ms,电流衰减呈现单指数衰减,衰减的时间常数为16.39±4.23ms。离子通道电导,在膜电位钳制于氯反转电位以下时为20.60±2.46 pS,反转电位以上时为50.83±8.51 pS。结论体外培养海马神经元的GABA-A受体全细胞离子通道平均电流随培养时间延长逐渐增加,到培养10时达最大,同时离子通道电导在不同钳制电位下显著不同。Objective: To study the electrophysiological characteristics of GABA-A receptor ion channel on cultured hippocampal neurons during their development. Methods: The patch clamp was used to record the changes in average whole cell current and the conductance of GABA-A receptor on cultured hippocampal neurons in different developing stages. Results: The whole-cell GABA-A receptor ion channel recurrent of cultured hippocampal neurons were 12.04±3.12 pA, 18.21±3.68 pA and 20.15±4.03 pA on the third, seventh and tenth in vitro days, and the increasing time of average whole cell current from 10% to 90% was 2.93±0.23 ms, while the current decay could be fit by one exponent with the time constant of 16.39±4.23 ms. The conductance of the channel remained constant in different developing stages but the channel conductance was variable to the holding potential, and with 20.60±2.46 pS at the membrane potential below the chlorine-inversion potential and 50.83±8.51 pS above the chlorine-inversion potential. Conclusion: The whole-cell recurrent of NMDA receptor ion channel on cultured hippocampal neurons increases with the in vitro time and the channel conductance remains steady in different developing stages but differs in membrane potential and the chlorineinversion potential.
